膀胱癌组织中CBX7 mRNA、miR-21表达及诊断价值分析

摘要:

目的 检测在膀胱癌组织中色素框同源蛋白7(CBX7 mRNA)、微小核糖核酸21(miR-21)的表达情况,并分析二者联合对膀胱癌的诊断价值。方法 收集2021年1月至2023年12月于西安高新医院进行手术治疗的120例膀胱癌患者的癌组织和癌旁组织样本,采用实时荧光定量聚合酶链反应法检测CBX7 mRNA和miR-21的表达,分析膀胱癌组织和癌旁组织中CBX7 mRNA和miR-21的表达情况、不同临床病理特征患者膀胱癌组织CBX7mRNA、miR-21表达情况,采用Pearson相关性分析法分析膀胱癌组织中CBX7 mRNA与miR-21表达的相关性,并采用受试者工作特征(ROC)曲线分析CBX7 mRNA、miR-21对膀胱癌的诊断价值。结果 膀胱癌组织中CBX7 mRNA表达水平显著低于癌旁组织(P<0.05),miR-21表达水平显著高于癌旁组织(P<0.05)。TMN分期为Ⅲ~Ⅳ期、低分化、有淋巴结转移的患者膀胱癌组织CBX7 mRNA表达低于TMN分期为Ⅰ~Ⅱ期、中高分化、无淋巴结转移的患者(P<0.05),TMN分期为Ⅲ~Ⅳ期、低分化、有淋巴结转移的患者膀胱癌组织miR-21表达高于TMN分期为Ⅰ~Ⅱ期、中高分化、无淋巴结转移的患者(P<0.05)。膀胱癌组织CBX7 mRNA与miR-21表达呈负相关(r=-0.188,P<0.05);CBX7 mRNA、miR-21联合诊断膀胱癌的曲线下面积为0.945,敏感度、特异度分别为96.67%、82.50%。结论 膀胱癌组织中CBX7 mRNA表达下调,miR-21表达上调,CBX7 mRNA与miR-21联合检测对膀胱癌的辅助诊断具有更高的价值。

Abstract:

Objective To investigate the expression of CBX7 and miR-21 in bladder cancer tissues, and analyze the diagnostic value of their combination in bladder cancer. Methods The cancer tissue samples and adjacent tissues of 120 bladder cancer patients who underwent surgical treatment in Xi 'an Gaoxin Hospital from January 2021 to December 2023 were collected. The expressions of CBX7 mRNA and miR-21 were detected by real-time quantitative PCR, the expressions of CBX7 mRNA and miR-21 in bladder cancer tissues and adjacent tissues were analyzed. Pearson method was used to analyze the correlation between the expression of CBX7 mRNA and miR-21 in bladder cancer tissues. The ROC curve was used to analyze the diagnostic value of CBX7 mRNA and miR-21 expression in patients with bladder cancer. Results The expression of CBX7 mRNA in bladder cancer tissue was significantly lower than that in adjacent tissue(P<0.05), and the expression of miR-21 was significantly higher(P<0.05); The expression of CBX7 mRNA in bladder cancer tissues of patients with TNM stage Ⅲ-Ⅳ, low differentiation and lymph node metastasis was lower than that of patients with TNM stageⅠ-Ⅱ, moderately and high differentiation and no lymph node metastasis. The expression of miR-21 in bladder cancer tissues of patients with TNM stage Ⅲ-Ⅳ, low differentiation and lymph node metastasis was higher than that of patients with TMN stageⅠ-Ⅱ, moderately and high differentiation and no lymph node metastasis(P<0.05); The expression of CBX7 mRNA and miR-21 in bladder cancer tissue was negatively correlated(r=-0.188, P<0.05); The area under the ROC curve of CBX7 mRNA combined with miR-21 to diagnose bladder cancer was 0.945, and the sensitivity and specificity were 96.67% and 82.50%, respectively. Conclusion The expression of CBX7 mRNA in bladder cancer tissue is down-regulated, and the expression of miR-21 is up-regulated. The combined detection of CBX7 mRNA and miR-21 is of high significance for the auxiliary diagnosis of bladder cancer patients.
